Study Suggests Ketogenic Diet May Enhance CAR T Cell Cancer Treatment
Researchers from the University of Pennsylvania’s Perelman School of Medicine and Penn Medicine’s Abramson Cancer Center have discovered that a ketogenic diet can potentially enhance the efficacy of CAR T cell therapy, a personalized cancer treatment. CAR T cell therapy, which reprograms patients’ immune cells to target and destroy cancer cells, has been a breakthrough in treating blood cancers. However, it doesn’t succeed for all patients.
The study, co-led by postdoctoral fellow Shan Liu and medical student Puneeth Guruprasad, explored the impact of various diets on the cancer-fighting abilities of CAR T cells in mice with diffuse-large B-cell lymphoma. Results demonstrated that mice on a ketogenic diet exhibited superior tumor control and survival compared to those on other diets. The team identified increased levels of beta-hydroxybutyrate (BHB), a metabolite rising from a ketogenic diet, as essential to enhancing CAR T cell function.
“Our research suggests that CAR T cells thrive on BHB more than glucose, enhancing their cancer-fighting power,” explained Guruprasad. Currently, a Phase I clinical trial at Penn Medicine’s Abramson Cancer Center is evaluating BHB’s potential to amplify CAR T cell therapy.
Dr. Maayan Levy, an assistant professor of Microbiology, remarked, “This intervention is cost-effective and likely low in toxicity. If clinical trials are successful, this approach could be combined with other treatments to bolster anti-cancer effects.”
The implications of this study are significant, presenting a promising avenue for improving cancer treatment outcomes through dietary modifications.
